Chymosin is an aspartic protease produced in the glandular stomach of adult ruminants and belongs to the pepsin family. Chymosin has been used in the industrial production of cheese and milk whey because it shows superior characteristics compared to other proteases. To examine the potential of chymosin as a biocatalyst, the gene encoding chymosin was expressed in two Escherichia coli expression systems, one in the presence and one in the absence of the chaperone DegP. E. coli strains containing chymosin could be selected by adding to the growth medium n-hexyl-beta-d-galactopyranoside and subsequently by screening for chymosin activity. Protein engineering was performed to optimize production in order to enhance the yield of correctly folded and active enzyme.
